Mechanostat function during skeletal development.

چکیده

these genes prevents development of skeletal muscle and mouse pups are born with a skeleton that lacks many of the features seen normally. Bones are straight instead of curved, and lack either the narrowing of the mid-shaft of the cortex or the flaring of the metaphyses seen in WT controls. Even more striking is the absence of the traction epiphyses, proving Julius Wolff’s assertion over 100 years ago that these develop because of muscular activity. MicroCT analysis of neonatal skeletons revealed reduced lengths of mineralized cortex, and low mineral density. These findings show that bone’s adaptive response to exercise begins in utero and that movement during skeletogenesis is a powerful influence on skeletal form at birth. If the fetal origins of adult disease hypothesis are applicable to bone, then the movements of the fetus may affect susceptibility to bone disease in later life some 50-60 years later. Hylonome

Influence of body weight on bone mass, architecture and turnover.

Weight-dependent loading of the skeleton plays an important role in establishing and maintaining bone mass and strength. This review focuses on mechanical signaling induced by body weight as an essential mechanism for maintaining bone health. In addition, the skeletal effects of deviation from normal weight are discussed. The magnitude of mechanical strain experienced by bone during normal acti...

Size, structure and gender: lessons about fracture risk.

The differences in age-related fracture risks among men and women must reflect gender differences in the relevant variables. We are concerned here with gender differences in structural variables that relate to the size and shape of bones. As children grow, their bones grow in diameter through periosteal modeling. Studies show that radial growth is driven by mechanical forces and is not just "ge...

Towards a cell-based mechanostat theory of bone: the need to account for osteocyte desensitisation and osteocyte replacement.

Bone׳s mechanostat theory describes the adaptation of bone tissues to their mechanical environment. Many experiments have investigated and observed such structural adaptation. However, there is still much uncertainty about how to define the reference mechanical state at which bone structure is adapted and stable. Clinical and experimental observations show that this reference state varies both ...

Exercise During Growth: Compelling Evidence for the Primary Prevention of Osteoporosis?

Exercise during growth is widely recommended as a key strategy in the primary prevention of osteoporosis. This theory seems reasonable if exercise is pursued into adulthood, but perhaps not if exercise is ceased; in the latter case, the mechanostat theory predicts a decrease in bone strength due to a decrease in the largest forces applied to the skeleton. However, recent findings suggesting mai...

Assessment of the influence of body composition on bone mass in children and adolescents based on a functional analysis of the muscle-bone relationship.

BACKGROUND The functional model of skeletal development considers the mechanical factor to be the most important skeletal modulant. The aim of the study was a functional analysis of the bone-muscle relationship in children with low and normal bone mass. MATERIAL AND METHODS The study involved 149 children with low and 99 children with normal bone mass (control group). All patients underwent a...
